Global transcriptional analysis of S. mutans sugar transporters

(Submitter supplied) The transport of carbohydrates by Streptococcus mutans is accomplished by the phosphoenolpyruvate-phosphotrasferase system (PTS) and ATP-binding cassette (ABC) transporters. To undertake a global transcriptional analysis of all S. mutans sugar transporters simultaneously, we used a whole-genome expression microarray. Global transcription profiles of S. mutans UA159 were determined for several carbohydrates. more...

Role of EIIABMan and EIIGlc in regulation of energy metabolism, biofilms, and competence in Streptococcus mutans

(Submitter supplied) The phosphoenolpyruvate:sugar phosphotransferase system (PTS) is the major carbohydrate transport system in oral streptococci. The mannose-PTS of Streptococcus mutans, which transports mannose and glucose, is involved in carbon catabolite repression (CCR) and regulates the expression of known virulence genes. In this study, we investigated the role of EIIGlc and EIIABMan in sugar metabolism, gene regulation, biofilm formation, and competence. more...

COORDINATED REGULATION OF THE EIIMAN AND FRURKI OPERONS OF STREPTOCOCCUS MUTANS BY GLOBAL AND FRUCTOSE-SPECIFIC PATHWAYS

(Submitter supplied) The glucose/mannose-PTS permease EIIMan encoded by manLMN in the dental caries pathogen Streptococcus mutans has a dominant influence on sugar-specific, CcpA-independent catabolite repression (CR). Mutations in manL affect energy metabolism and virulence-associated traits, including biofilm formation, acid tolerance and competence. Using promoter:reporter fusions, expression of the manLMN and the fruRKI operons, encoding a transcriptional regulator, a fructose-1-P kinase and a fructose-PTS permease EIIFru, respectively, was monitored in response to carbohydrate source and in mutants lacking CcpA, FruR, and components of EIIMan. more...

Regulation of PTSBio by transcriptional repressor NigR

(Submitter supplied) In this study we analyzed the transcriptional regulation of the operon (OBio) encoding the PTSBio and showed that it was repressed by NigR, a LacI-like transcriptional regulator. To elucidate the role of NigR in regulation of S. mutans carbohydrate transporters we performed full genome expression microarrays using custom Affymetrix chip. Expression microarray analysis was performed with ΔnigR grown in CDM supplemented with glucose and compared to that of WT grown under identical conditions. more...

The Role of CcpA in Gene Regulation in Streptococcus mutans

(Submitter supplied) CcpA is a global regulator of transcription in Gram-positive bacteria that controls gene expression in response to carbohydrate availability. Using the fructan hydrolase (fruA) gene of S. mutans as a model, we demonstrate that CcpA does indeed play a major role in carbohydrate catabolite repression. Using DNA microarrays, the expression of at least 170 genes differed in CcpA-deficient cells compared to the parental strains when cells are grown in the presence of glucose, but only 90 genes showed altered expression when cells were cultivated in the poorly-repressing substrate galactose. more...

Sucrose- and fructose-specific effects on the transcriptome of Streptococcus mutans probed by RNA-Seq

(Submitter supplied) For Streptococcus mutans, the major etiologic agent of human dental caries, sucrose is the carbohydrate that contributes in the most significant manner to establishment, persistence and virulence of the organism. However, because this organism produces multiple extracellular sucrolytic enzymes that can release hexoses from sucrose, it has not been possible to study the specific effects of sucrose transport and metabolism on gene expression in the absence of carbohydrates that by themselves can elicit catabolite repression and induce expression of multiple genes. more...

Csac growth on model substrates found in lignocellulose

(Submitter supplied) The co-utilization of hexoses and pentoses derived from lignocellulose is an attractive trait for in microorganisms considered for consolidated biomass processing to biofuels. This issue was examined for the H2-producing, extremely thermophilic bacterium Caldicellulosiruptor saccharolyticus growing on oligosaccharides (composed of glucose and xylose) compared to monosaccharides. Based on the whole-genome transcriptional response analysis and comparative genomics, carbohydrate specificities for transport systems could be proposed for a number of the 24 putative carbohydrate ATP-binding cassette (ABC) transporters in the C. more...

Differential gene expression in Roseburia inulinivorans grown on inulin and starch

(Submitter supplied) Roseburia inulinivorans is a recently identified motile representative of the Firmicutes that contributes to butyrate formation from a variety of dietary polysaccharide substrates in the human large intestine. Microarray analysis was used here to investigate substrate-driven gene expression changes in R. inulinivorans A2-194. A cluster of fructo-oligosaccharide (FOS)/inulin utilisation genes induced during growth on inulin included one encoding a b-fructofuranosidase protein that was prominent in the proteome of inulin-grown cells. more...
