A DNA microarray was designed and constructed based on 2793 protein-coding sequences in the C. saccharolyticus genome obtained from the Department on Energy's Joint Genomes Institute (http://genome.ornl.gov/microbial/csac). OligoArray 2.0 (Rouillard, 2003) was used to generate one 60-mer oligonucleotide probe sequence for each open reading frame. The probes were synthesized (Integrated DNA Technologies, IA), re-suspended in 50% DMSO, and printed onto Ultragap microarray slides (Corning, NY) using a QArrayMini arrayer (Genetix, UK). Each probe was spotted five times onto each array to fortify statistical analysis.
