Title |
SK-N-MC_shPRC1_cds_dox+, 1 biologial Rep, 60h |
Sample type |
RNA |
Source name |
SK-N-MC_shPRC1_cds
Organism |
Homo sapiens |
Characteristics |
cell line: SK-N-MC cell type: Ewing sarcoma cell line genotype/variation: shRNA against PRC1 CDS region treatment: dox (1μg/ml)
Treatment protocol |
Cells were treated with/without dox (1μg/ml) for 60h, with a refreshment after 48h.
Growth protocol |
700.000 cells per flask/T25 flask were cultured for 60h with/without addition of dox (1μg/ml).
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted by using RNA isolation kit from Macherey-Nagel.
Label |
Biotin
Label protocol |
Biotinylated cRNA were prepared using Affymetrix GeneChip® WT PLUS Reagent Kit according to its manufacture's instruction from 6 μg total RNA.
Hybridization protocol |
Following fragmentation, 10 μg of cRNA were hybridized for 16 hr at 45C on Affymetrix Human Clariom D™ Arrays. Microarrays were washed and stained with Affymetrix GeneChip® 3000 Fluidics Station.
Scan protocol |
Microarrays were scanned with Affymetrix GeneChip® 3000 Scanner.
Description |
19059-0020
Data processing |
The data were analyzed with RMA using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100. annotation file: Clariom_D_Human.r1.na36.hg38.a1.transcript.csv
