H3K27me3 ChIP-seq experiment in fruitfly testis somatic gonadal cells

(Submitter supplied) In many metazoans, germ cells are separated from somatic lineages early in development and maintain their identity throughout life. Here we show that a Polycomb group (PcG) component, Enhancer of Zeste [E(z)] H3K27me3-specific methyltransferase, maintains germline identity in Drosophila adult testes. E(Pc) ChIP-seq experiment in fruitfly testis somatic gonadal cells and RNA-seq experiment in fruitfly testis

(Submitter supplied) With the ChIP-seq experiment, we identified direct target genes of E(Pc) specifically in somatic gonadal cells. We found that the E(Pc)-binding genes are enriched with signaling pathway components, consistent with our functional data showing prevailing germline defects even when E(Pc) function is exclusively compromised in somatic gonadal cells. In addition, we also performed RNA-seq to compare transcriptomes between E(Pc) knockdown testes and control testes.

Piwi is a key regulator of both somatic and germline stem cells in the Drosophila testis

(Submitter supplied) The Piwi-piRNA pathway is well known for its germline function, yet its somatic role remains elusive. We show here that Piwi is required autonomously not only for germline stem cell (GSC) but also for somatic cyst stem cell (CySC) maintenance in the Drosophila testis. Neonatal and adult human testis defined at the single-cell level

Comparison of testis transcriptome changes resulting from mutations in dany and representative meiotic arrest genes

(Submitter supplied) The Drosophila gene dany is expressed specifically in spermatocytes and encodes a nuclear protein with similarity to the products of the genes distal antenna (dan) and distal antenna-related (danr). Loss of dany function results in male sterility. Mutant spermatocytes fail to differentiate similar as previously observed in mutants lacking tTAF and tMAC function. Jarid2 methylation is required for PRC2 functioning during ES cells differentiation

(Submitter supplied) We report that Jarid2 is methylated at K116 by the polycomb complex PRC2. We analyzed Jarid2 versus Methylated Jarid2 genomic localization by ChIP-seq. We also study H3K27 enrichment in Jarid2 knock out ES cells and in cells rescued for Jarid2 or a mutant that cannot be methylated. ChIP-seq for H3K27me3 was perfomed in undiffrentiated ES cells as well as after 8 days of diffrentiation toward Embroyid Bodies (Hanging Drop method).
