Extraction protocol |
The heart, quadriceps femoris, and diaphragm were dissected and flash frozen in liquid nitrogen-cooled isopentane. Samples were homogenised using Precellys 24 (Bertin Technologies, France), and RNA was extracted using TRIzol reagent (Invitrogen) as according to the manufacturer’s instructions. Each RNA sample was analysed using the Agilent 2100 Bioanalyzer (Agilent Technologies, USA) to assess RNA quality.

1 ug of total RNA was labeled using the FlashTag Biotin HSR kit (Genisphere).

Total labeled RNA was hybridised to the Affymetrix GeneChip miRNA array 2.0 using the manufacturer's protocols.

Image files from scanning (.dat and .cel files) were generated with the Affymetrix Expression Console package.

Quad_C57_4 miRNA expression data from diaphragm, heart and quadriceps of mdx and WT C57/B10 mice.

Data processing |
The processing and data analyses were performed in Affymetrix miRNA QCTool, as well as in DNA-Chip Analyzer (dChip) and Excel. The analysis was done according to the following workflow; BC-CG background adjustment, Quantile normalisation and Median Polish summarisation. Since the miRNA 2.0 array covers 131 organisms, we sorted out Mus musculus transcripts only.
