Growth protocol The animals were bleached to obtain eggs, which were hatched in M9 overnight. The next day, L1s were seeded onto NGM plates with K12 WT bacteria and grown until L4 stage before being harvested.

Extraction protocol The L4 animals were washed with water and the worm pellet was extracted using TRIzol reagent (Invitrogen). The library was constructed by BGI using the standard DNBSEQ Eukaryotic Strand-specific Transcriptome Resequencing service (P_Stranded Library & PE100 Seq).

Data processing The trimmed sequencing reads were mapped to C. elegans reference genome (WB235) using STAR (2.7.10) with the option --quantMode geneCounts. The counts were then normalized and the differentially expressed genes were then identified using the standard pipeline of DESeq2 in R.
