Extraction protocol |
Synovium samples from four RA patients were obtained. Single-cell suspensions were prepared as previously described (PMID: 29996944). CD4+ T cells were magnetically sorted by MojoSort Mouse CD4 T Cell Isolation Kit, and susequently CD3+ CD4+ T cells were sorted by a FACS sorter. Sorted cells were immediately subjected to 10X Chromium controler to prepare libraries. Libraries were prepared using Chromium Next GEM Single Cell 5'Kit v2 and Single-Cell Human TCR Amplificication Kit according to the protocol.
