Title |
Yeast_DMSO_30min_rep1 |
Sample type |
RNA |
Source name |
S. cerevisiae, DMSO-treated
Organism |
Saccharomyces cerevisiae |
Characteristics |
strain: BY4741
Treatment protocol |
Following 30 min small molecule treatment, cells were harvested and flash frozen in liquid nitrogen.
Growth protocol |
Yeast cells were grown to mid-log phase (0.5–2 x 107 cells/ml) at 30°C, 300 rpm, in YPD medium before treatment with small molecules for 30 min.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated using the RiboPure™-Yeast kit (Ambion) and RNA quality was checked using Agilent 2100 Bioanalyzer (Agilent Technologies, Palo Alto, CA).
Label |
Biotin
Label protocol |
Biotin-labeled cRNA probes were generated from total RNA using the One-Cycle Target Labeling Assay (Affymetrix).
Hybridization protocol |
Hybridization of labeled cRNA probes to Affymetrix GeneChip Yeast Genome 2.0 arrays (www.affymetrix.com) was done according to manufacturer’s specifications.
Scan protocol |
The arrays were scanned using Affymetrix GeneChip Scanner 3000 and raw data generated using the GeneChip Operating System GCOSv1.4.
Description |
growth in YPD media
Data processing |
Gene expression data were normalized in dChip (www.dchip.org). Model-based expression indices (MBEIs) were calculated and log2 transformed. We have only shown the data pertaining to the Saccharomyces cerevisiae probes and had deleted the data pertaining to Saccharomyces pombe since that data is irrelevant to our study, which only uses S. cerevisiae. We do show ALL Saccharomyces cerevisiae probe data.
