Treatment protocol Adult female Sprague-Dawley rats (220-250 g) underwent surgical T10 laminectomy and moderate contusive injury (175 kDyn) by an Infinite Horizons impactor. Treatment groups were acutely administered the AAV5-GfaABC1D-dYFP reporter vector at the time of surgery by stereotaxic microinjection. Following recovery, at 7- and 28-days post-injury animals were euthanised, perfused with ice-cold saline and a 5 mm region centered on the spinal lesion excised to be snap frozen.

Extracted molecule total RNA
Extraction protocol Total RNA was extracted by mechanical tissue dissociation, TRIzol / chloroform extraction and phase separation. RNA extract quantified by Implen NP80 nanophotometer and quality assed by Agilent Bioanalyzer (RIN >8.80).
Label Biotin
Label protocol Biotinylated cDNA were prepared according to the standard Affymetrix protocol from 200 ng total RNA.
 
Hybridization protocol Rat Clariom S arrays were incubated in the Affymetrix® GeneChip® Hybridization Oven 645 at 45° C / 60 RPM for 16-18 hours.
Scan protocol GeneChips were scanned with the GeneChip™ Scanner 3000 (Applied Biosystems, #00-0186) according to manufacturer's instructions.

Data processing Raw intensity CEL files were imported into Transcriptome Analysis Console (TAC software v4.0.2.15. Applied Biosystems), and gene level expression analysis was performed using the default ANOVA settings.
