Title |
F114#1_1_A8916 |
Sample type |
RNA |
Source name |
spleen
Organism |
Mus musculus |
Characteristics |
tissue: spleen Sex: female strain: C57BL/6 parasite strain: F114#1 replicate mouse: 1 rna sample id: A8916
Treatment protocol |
An inoculum containing 10×106 iRBC suspended in 100 ml of phosphate-buffered saline, pH 7.4, from donor mice was injected intravenously into each experimental C57BL/6 mouse to initiate infection. Naive mice receiving an equivalent number of uninfected RBCs served as a negative control group. Three to six mice as a group were infected with each parasite, and parasitemia were monitored by microscopic examination of Giemsa-stained thin tail blood smears.
Growth protocol |
Inbred female C57BL/6 mice, aged 6–8 weeks, were purchased from Charles River Laboratory.
Extracted molecule |
polyA RNA |
Extraction protocol |
RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyzer or TapeStation.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared with the Ambion TotalPrep kit for Illumina arrays
Hybridization protocol |
Standard Illumina hybridization protocol
Scan protocol |
Standard Illumina scanning protocol using HiScan-SQ
Description |
spleen RNA 24 hr post infection with strain F114#1,mouse replicate 1, RNAsampleID=A8916
Data processing |
Raw data filtered to include only probes detected in at least two arrays, using the detection_pvalue parameter output from Genome Studio, cutoff is 0.1; Each array normalized using quantile normalization of bead_average intensity, performed using JMP/Genomics software.
