Total RNA were isolated from paraffin-embedded human colorectal cancer tissues using mirVanaTM RNA Isolation Kit (AM1561) according to the manufacturer's instructions.

Total RNA were dephosphorylated, denaturated, and then labeled with Cyanine3-pCp.

Hybridization protocol |
The labeled RNA were hybridized for 20 hr at 55°C on Agilent-038166 Array. After hybridization, microarrays were washed 5 minute at room temperature with GE Wash Buffer 1 (Agilent) and 5 minute with 37°C GE Wash buffer 2 (Agilent).

Scan protocol |
After washing, the arrays were scanned with the Agilent Scanner G2505C (Agilent Technologies).

Data processing |
Feature Extraction software (version10.7, Agilent Technologies) was used to analyze array images to get raw data. Next, Genespring software (version 10.0; Agilent Technologies) was employed to finish the basic analysis with the raw data according to scale to mean of all samples. Differentially expressed miRNAs were identified through fold change as well as P value calculated using t-test.
