Growth protocol |
Seeds were surface sterilized with 2% sodium hypochlorite and 0.01% triton X-100 for 5 min followed by rinsing in sterile water for 5 times. Seeds were then suspended in 0.1% agar solution and dispensed on 0.8% agar-gelled MS medium containing 2% sucrose. Plates were sealed and shifted to cold room for 72 h to break dormancy and facilitate uniform germination and then transferred to culture room under continuous illumination.

Biotinylated cRNA was prepared according to the standard Affymetrix protocol from 5 ug total RNA (Expression Analysis Technical Manual, Affymetrix).

Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45 degree celsius on Affymetrix ATH1-121501 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450 using script EukGE-WS2v4.

Description |
Arabidopsis 10-day-old wild-type seedlings grown under normal growth conditions were used for total RNA isolation. The high-quality total RNA (260/280 ratio between 1.9-2.1 and 260/230 ratio >2.0) was used for microarray analysis
