Extraction protocol |
RNA was extracted from mouse mammary tumors and normal mammary glands that had been snap-frozen immediately following animal necropsy. Tissues were powdered under liquid nitrogen, homogenised in Qiashredder columns and RNA isolated using the miRNeasy Mini Kit (Qiagen).

Labeling was carried out with the miRNA Complete Labeling and Hyb Kit and microRNA Spike-in kit (both Agilent).

Hybridization protocol |
miRNA was hybridized to Agilent 8x60K miRNA Oligo Microarrays (G4872A-038112). Arrays were washed as directed by the manufacturer.

Scan protocol |
Agilent dual laser scanner (P/N: G2505B). Feature extraction was carried out using Agilent software (FE 10.7.3).

Data processing |
Raw Cy3 intensities were background corrected using the RMA background correction algorithm. Data was log2 transformed and quantile normalized.
