Overall design |
After 5 days of RNAi, total RNA was harvested using Trizol reagent from 5 x 106 S2 cells, and mRNA was purified using oligo-dT-conjugated magnetic beads. Library construction was conducted using TruSeq RNA sample preparation kits (Illumina). All sequencing data were obtained from the Vincent J. Coates Genomics Sequencing Laboratory at UC Berkeley.
