Title Comparative analysis of mouse CD8+ Teff cells infiltrating distinct organ targets of aGVHD by gene expression profiling

Summary This study seeks a better understanding of the functional differences emerging between mouse CD8+ T effector (Teff) cells infiltrating three distinct mucosal environments; the small intestine, the lung and the liver. CD8+ Teff cells immigrating in the target organs were generated using a murine experimental acute graft versus host disease (aGVHD) model. For this purpose, congenially labeled CD45.1 OT-I CD8+ Teff cells were transplanted to and activated in a lethally irradiated allogeneic CD45.2 host constitutively expressing their cognate antigen, the chicken ovalbumin protein. CD8+ Teff cells were retrieved from their respective organ environments by automated organ disintegration and MACS sorting, and compared with each other by whole genome gene expression profiling. Peripheral blood derived circulating CD8+ Teff cells were also analyzed, serving as circulating peripheral T effector cell controls
 
Overall design Lethal acute graft versus host disease was induced in healthy 8-16 weeks old C57Bl/6 m-act OVA female mice (n=5-6) by allogeneic bone marrow transplantation. Recipient m-act OVA mice received total body irradiation and were transplanted with 3x10E+6 bone allogeneic marrow cells and 1.5x10E+7 splenocytes from 8-16 weeks old C57Bl/6 OT-I mice serving as donors. Graft OT-I CD8+ T cells were obtained from CD45.1 animals, while all other graft cells and recipients carried the CD45.2 allele. Four days after bone marrow transplantation grafted CD45.1 CD8+ OT-I T cells were recovered from the livers, lungs and small intestines of m-act OVA mice with full-fledged acute GVHD by MACS sorting of CD45.1+ cells, and subjected to whole genome gene expression profiling.
