Overall design |
Total bone marrow was cultured in 40 ng/mL GM-CSF enriched RPMI-media for 7 days. A fraction of the floating cells were stimulated with 100 ng/mL LPS for 16 h, after with both unstimulated and stimulate DC were sorted by FACS for DC markers and activation markers (CD86). Unactivated DC from unstimulated samples (CD86-) were isolated together with activated (adherent) DC from LPS stimulated samples (CD86+), and total RNA was isolated.
