Title |
miRNA profiling in radiated mice bone marrow and hypothalamus pretreated with radioprotectants. |
Organism |
Mus musculus |
Experiment type |
Non-coding RNA profiling by array
|
Summary |
To further understanding of miRNA expression during radioprotectation, we used microarray analysis to determine the differential expression of miRNAs in the bone marrow of mice exposed to high-dose IR with and without pretreatment with three types of radioprotectants: Amifostine (WR2721), CBLB502, and Nilestriol, as well in the hypothalamus of mice exposed to IR with and without pretreatment with amifostine. Overall, amifostine treatment led to a similar expression pattern of miRNAs to that of CBLB502 but not to that of nilestriol in bone marrow. The radioprotectants exhibited mostly opposite effects against IR-induced miRNAs. These results expand the biological function of miRNAs in mediating the activity of radioprotectants, further offering insights into the molecular mechanism underlying radioprotection.

Overall design |
Radiation induced miRNA expression in mice bone marrow and hypothalamus pretreated with amifostime, CBLB502 and nilestriol was measured at 12 hours after exposure to doses of 8 Gy gamma-rays. The bone marrow from three animals were pooled in one sample for each treatment.
