Overall design We performed RNA-seq and small-RNA seq on human ESCs differentiated into β like cells in vitro, same cells transplanted into kidney capsule of mouse recipients, and fresh human islets. We also performed RNA-seq on hESCs genomically modified with dox inducible CRISPR knockout of LIN28 and then differentiated into β like cells plus/mius doxycycline
