Neuronal DNA damage response-associated dysregulation of signalling pathways and cholesterol metabolism at th earliest stages of Alzheimer-type pathology

(Submitter supplied) High levels of oxidative stress and an associated neuronal DDR occur at the earliest stages of Alzheimer pathology (low Braak stage), and is associated with cognitive impairment. The aim of the present study was to combine immuno-LCM and microarray analysis to characterize the neuronal transcriptome at low Braak stages (Braak 0-II), with respect to the neuronal DDR, in post-mortem human frontal cortex derived from the Medical Research Council Cognitive Function and Ageing Study (MRC-CFAS).

The reverse transcriptase inhibitor 3TC protects against age-related cognitive dysfunction

(Submitter supplied) Aging is the primary risk factor for most neurodegenerative diseases, including Alzheimer's disease. Major hallmarks of brain aging include neuroinflammation/immune activation and reduced neuronal health/function. These processes contribute to cognitive dysfunction (a key risk factor for Alzheimer's disease), but their upstream causes are incompletely understood. Age-related increases in transposable element (TE) transcripts might contribute to reduced cognitive function with brain aging, as the reverse transcriptase inhibitor 3TC reduces inflammation in peripheral tissues and TE transcripts have been linked with tau pathology in Alzheimer's disease. more...

Gene expression in primate postnatal brain through lifespan - cerebellar cortex

(Submitter supplied) We investigated molecular changes during human, chimpanzee, and rhesus macaque postnatal brain development at the transcriptome, proteome, and metabolome levels in two brain regions: the prefrontal cortex (PFC) that is involved in several human-specific cognitive processes, and the cerebellar cortex (CBC) that may be functionally more conserved. We find a nearly three-fold excess of human-specific gene expression changes in PFC compared to CBC. more...
