Transforming growth factor beta effect on cytokine-activated, NOD-derived, splenic natural killer cells

Analysis of NOD-derived, splenic natural killer (NK) cells cultured with or without cytokines interleukin (IL)-12 + IL-18 in the absence of TGF-β, or with both cytokines in the presence of TGF-β. Results provide insight into the impact of TGF-β on NK cells independent of general activation.

Gene expression profiling of NK cells isolated from the pancreas of BDC TCR expressing NOD mice before or after Treg ablation by DTR. Also, NK cells cultured in vitro with activating cytokines IL12 and IL-18 with or without regulatory TGFb.

(Submitter supplied) Following Treg ablation in the BDC/NOD.Foxp3-DTR strain, NK cells produce IFNg and accumulate to higher percentage and number. We explored the signature pathways responsible for this phenomenon using microarray prolifing and comparison to other activation signatures.

Profiling of aggressive vs benign pancreatic infiltrates in the BDC2.5 Tg model of Type I diabetes

(Submitter supplied) This study was performed to understand what controls the aggressivity of the pancreatic infiltrate during type-I diabetes development. We used the BDC2.5 transgenic mouse model. Samples were obtained at the age of onset of insultis. Depending on their genetic background, mice transgenic for the BDC2.5 T cell receptor present very different forms of insulitis. Insulitis: comparison of innocuous and destructive forms

Analysis of CD4+ automimmune T and CD45+ hematopoeitic cells from non-obese diabetic (NOD) and C57BL/6-H-2 g7 transgenics with the BDC2.5 T cell receptor (TCR) from a diabetogenic T cell. BDC2.5 TCR in NOD and in C57BL/6-H-2 g7 results in innocuous and destructive insulitis respectively.
