Multiparameter functional diversity of human C2H2 zinc finger proteins [RNA-Seq]

(Submitter supplied) C2H2 zinc finger proteins represent the largest and most enigmatic class of human transcription factors. Their C2H2 arrays are highly variable, indicating that most will have unique DNA binding motifs. However, most of the binding motifs have not been directly determined. We have determined the binding sites and motifs of 119 C2H2 zinc finger proteins and the expression pattern of 80 cell lines overexpressing C2H2 zinc finger proteins in order to study the role of C2H2 zinc finger proteins in gene regulation.

The C2H2-ZF transcription factor Zfp335 recognizes two consensus motifs using separate zinc finger arrays

(Submitter supplied) The complexities of DNA recognition by transcription factors with multiple Cys2-His2 zinc fingers (C2H2-ZFs) remain poorly studied. We previously reported a mutation (R1092W) in the C2H2-ZF transcription factor Zfp335 which led to selective loss of binding at a subset of targets, although the basis for this was unclear. Genome wide analyses of Su(Hw)M393 binding in the Drosophila ovary

(Submitter supplied) The Suppressor of Hairy-wing [Su(Hw)] protein is a twelve zinc-finger DNA binding transcriptional regulator. Here, we map the genome wide association of Su(Hw) protein expressed from the novel su(Hw)M393 allele, which encodes a protein with a C350S substitution that inactivates ZF4. These analyses demonstrate that ZF4 is required for association with a subset of Su(Hw) binding sites and suggests that these sites may be primarily associated with insulator function.
