Gene expression analysis in human colon cancer cells and liver cancer cells with FoxA knockdown.

(Submitter supplied) FoxA transcription factors are involved in development and tumorigenesis of the gastrointestinal tract. However, the downstream programs controlled by FoxA factors remain poorly understood. The goal of this study is to understand the transcriptional responses regulated by FoxA proteins in liver and colon cancer cells.

Transcriptional responses of human colon and liver cancer cells to TCF inhibition.

(Submitter supplied) Dysregulation of Wnt/TCF signaling is closely associated with cancers arising from the gastrointestinal tract, inlcluding colon cancer and liver cancer. The goal of this study is to understand the transcriptional programs underlying Wnt/TCF activation in gastrointestinal cancers. We examined the transcriptional responses to TCF inhibition in cultured human colon cancer cells and liver cancer cells that are characteristic of Wnt pathway activation.

Delineation of EZH2 oncogenic functions in hepatocellular carcinoma

(Submitter supplied) The goal of this study was to delineate the important EZH2 direct target genes that mediate the oncogenic properties of EZH2 in HCC. The EZH2 direct target genes in two HCC cell lines were identified by chromatin immunoprecipitation microarray (ChIP-chip) analysis and later confirmed by independent ChIP-PCR. The functions of the target genes were further examined.

ERG is a critical regulator of Wnt/LEF1 signaling in prostate cancer

(Submitter supplied) Chromosomal translocations juxtaposing the androgen-responsive TMPRSS2 promoter with the ETS-family transcription factor ERG result in aberrant ERG up-regulation in approximately 50% of prostate cancers. Studies to date have demonstrated important roles of ERG in inducing oncogenic properties of prostate cancer. Its molecular mechanisms of action, however, are yet to be fully understood. To address these questions, we generated engineered LNCaP cells with ERG overexpression followed by LEF1 knockdown as well as control cell lines. more...

FGF Receptor and Wnt-1 Signaling Cooperate in Mammary Tumorigenesis

(Submitter supplied) The fibroblast growth factor pathway is known to cooperate with the highly oncogenic Wnt/beta-catenin pathway in mouse models of breast cancer. To investigate the mechanisms involved in this cooperativity, we utilized MMTV-driven transgenic mouse lines expressing a drug-inducible model for FGF Receptor signaling (iFGFR) crossed with the previously characterized MMTV-Wnt-1 mouse model. In these bigenic mice, both iFGFR1 and iFGFR2 activation resulted in a dramatic enhancement of mammary tumorigenesis. more...

TNFR2 HnRNPK-YAP Signaling Axis Promotes Primary Liver Cancer Development in Hepatic Progenitor Cells

(Submitter supplied) We performed single-cell RNA sequencing (scRNA-seq) analysis using clinical samples of PLC patients. We first generated scRNA-seq profiles of freshly isolated PLC from patients with 10x Genomics. A total of 25,189 single-cell transcriptomes from tumors of two patients were obtained after initial quality controls.

Wnt/β-catenin signaling regulates amino acid metabolism through the suppression of CEBPA and FOXA1 in liver cancer cells [Array]

(Submitter supplied) In this study, we showed that activated Wnt/β-catenin signaling suppressed HAL and ARG1, genes associated with cellular metabolism, via CEBPA and FOXA1 transcription factors. In contrast, inhibition of the Wnt signaling pathway increased HAL and ARG1, resulting in decreased intracellular levels of histidine and arginine in liver cancer cells. These data gain an insight into a new role of the Wnt signaling pathway in amino acid metabolism in liver cancer.
