Expression data from an involution time course in wild type and E2F3 +/- mice.

(Submitter supplied) We have compared an involution timecourse for wild type and E2F3 heterozygous mice on the Operon 3.0 platform. Three mice were taken for each genotype at each timepoint (Involution day 0, 1, 2, 3 and 4) and RNA was prepared from the mammary glands. Mouse Mammary Gland Development

(Submitter supplied) The mammary gland develops mainly postnatally, when during pregnancy the epithelium grows out into the mammary fat pad and forms a network of epithelial ducts. During pregnancy, these ducts branch and bud to form alveoli. These alveoli produce the milk during lactation. After 7 days of lactation, involution was induced by force weaning the pups. Efficient Mammary Gland Involution Requires c-Jun N-terminal kinase

(Submitter supplied) Involution returns the lactating mammary gland to a quiescent state after weaning. The mechanism of involution involves collapse of the mammary epithelial cell compartment. To test whether the cJUN NH2-terminal kinase (JNK) signal transduction pathway contributes to involution, we established mice with JNK deficiency in the mammary epithelium. We found that JNK is required for efficient involution. JNK deficiency did not alter the STAT3/5 or SMAD2/3 signaling pathways that have been previously implicated in this process. Nevertheless, JNK promotes the expression of genes that drive involution, including matrix metalloproteases, cathepsins, and BH3-only proteins. These data demonstrate that JNK has a key role in mammary gland involution post lactation.

Gene expression in mammary glands of lactating wild-type or Th-POK-deficient mice.

(Submitter supplied) The Th-inducing POK (Th-POK, also known as cKrox or ZBTB7B) transcription factor is a key regulator of lineage commitment of immature T cell precursors. It is yet unclear the physiological functions of Th-POK besides helper T cell differentiation. We found that Th-POK is restrictedly expressed in the luminal epithelial cells in the mammary glands. Loss of Caveolin-3 Induces the Development of a Lactogenic Microenvironment

(Submitter supplied) Here, we show that functional loss of a single gene is sufficient to confer constitutive milk protein production and protection against mammary tumor formation. Caveolin-3 (Cav-3), a muscle-specific caveolin-related gene, is highly expressed in striated and smooth muscle cells. We demonstrate that Cav-3 is also expressed in myoepithelial cells within the mammary gland. Caveolin-3 deficiency effect on virgin mammary glands

Analysis of caveolin-3 (Cav-3) deficient mammary glands from virgin females. Cav-3 deficient virgin mammary glands develop lobulo-alveolar hyperplasia, akin to the changes normally observed during pregnancy and lactation. Results provide insight into the molecular basis of this Cav-3 null phenotype.

Gene expression in bovine mammary gland in response to increased milking frequency as determined by microarray and SAGE

(Submitter supplied) Transcript profiling was performed by Affymetrix microarray analysis and SAGE to characterize changes in gene expression in the bovine mammary gland in response to 4× versus 2× daily milking during the first week of lactation. These changes in gene expression may contribute to the increased milk production observed in response to increased milking frequency. Our results indicate that increased milking frequency alters expression of genes in the mammary gland related to increased extracellular matrix remodeling, neovascularization, metabolism, cell proliferation and apoptosis. more...
