Study: RNA-sequencing, sporangium formation of Actinoplanes missouriensis

RNA-sequencing was conducted in the rare actinomycete Actinoplanes missouriensis to analyze gene expression profiles during sporangium formation. The bacterium forms motile zoospore-containing sporangia upon the substrate mycelia under adverse conditions. In this project, A. missouriensis was cultured on a sporangium-forming HAT agar medium for 1, 3, 6 and 40 days. At day 1, A. missouriensis grows as substrate mycelia. At day 3, sporangium formation starts. At day 6, a certain amount of sporangia are formed. And at day 40, much of the sporangia are supposed to be dormant. Total RNAs were extracted at each time point, and RNA-sequencing using Illumina HiSeq2500 sequencer was conducted.
