Treatment protocol |
Prior to treatment, cells were trypsinized, counted and seeded in 60mm cell culture dishes. 24 hours after cell seeding, the cell culture medium of the control group was replaced with fresh medium without TGF-beta, while the cell culture medium of the two TGF-beta treatment groups was replaced with fresh medium containing either 5ng/mL of TGF-beta-1 or TGF-beta-2. At 48 hours post treatment, cells were harvested and total RNA was extracted.

Growth protocol |
The NOF151 ovarian fibroblast cell line was cultured in a humidified incubator at 37C and 5% carbon dioxide. Culture medium M105/199 supplemented with 10% Fetal bovine serum and 1ng/mL Epidermal growth factor was used as growth medium.

Total RNA extraction was performed using the Purelink RNA Mini Kit (Invitrogen) according to the manufacturer's instructions.

Biotinylated cRNA were prepared using the MessageAmp Premier RNA Amplification Kit (Ambion) according to the manufacturer's instructions from 100ng total RNA.

Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Human Genome U133 Plus 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000 7G.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as the normalization method.
