Cells were cultured in Iscove's modified Dulbecco's medium (IMDM) supplemented with 10% (FBS) and 1% penicillin/streptomycin. TRAIL-resistant Jurkat clones (TR1, TR2, TR3) were established by selective pressure of TRAIL 1000 ng/mL over the period of 12 weeks.

Extraction protocol |
Total cellular RNA was isolated from biologic duplicates of untreated Jurkat cells (WT) and TRAIL resistant Jurkat cell line subclones (TR1, TR2, TR3) using RNAblue (Top-Bio, Czech Republic) according to the manufaccturer’s instructions. RNA quality was assessed using the Agilent 2100 Bioanalyzer (Agilent Technology). Extracted RNA was amplified using the Illumina RNA Amplification Kit (Ambion) and hybridized to the Illumina HumanRef-8 BeadChip (Illumina).
