Extraction protocol |
A total of 30ml blood was drawn and subjected to Ficoll seperation to obtain PBMCs. CD14+ monocytes were isolated from PBMC with CD14-conjugated magnetic bead. Total RNA was isolated from 1-2x10^6 monocytes with Qiagen Rneasy mini kit.

Half microgram of total RNA was amplified and labelled with Ambion MessageAmp™ II-Biotin Enhanced Single Round aRNA Amplification Kit (AM1790) using biotin-UTP.

Hybridization protocol |
Biotin labeled fragmented aRNA was sealed in the chamber of the microarray slide. Hybridized for 17 hours at 37C on a horizontal rocking platform at 300 rmp. The slides were then washed and stained with Steptavidin Alexa Fluor-647. The slides were washed after staining.

Scan protocol |
The arrays were then scanned on a Axon GenePix 4000B Scanner with PMT 600, single color.
