Treatment protocol PMNs (1 × 107) in RPMI/H were cultured with 1 nM PVL or iPVL at 37°C for 30, 60, or 180 min.
Growth protocol PMNs were isolated from venous whole blood of healthy individuals as described [PMID:11983860]. Each subject gave informed consent prior to participation in the study and all work was approved by the Institutional Review Board for Human Subjects, National Institute of Allergy and Infectious Diseases, National Institutes of Health, USA.

Extraction protocol At each indicated time point, PMNs were lysed with RLT buffer and RNA was purified
Label biotin
Label protocol 12 µg of biotin-labeled cRNA target as previously described [PMID:11983860]
 
Hybridization protocol Samples from 3 different PMN donors were hybridized on HU133+2 GeneChips (Affymetrix, Santa Clara, CA) according to manufacturer's recommendations
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 7GPlus.
