Treatment protocol Fathers fed on HFD vs. control were mated with females on normal chow. The female offspring's fat transcriptomics were analyzed through microarray analysis.
Growth protocol At 15 weeks of age, fathers were mated with normal females consuming chow to generate the F1 offspring.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions (Invitrogen).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45°C on GeneChip Rat Gene ST 1.0 arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using the Affymetrix Scanner GC3000.
Description Gene expression of fat from female offspring of fathers fed high-fat chow.
Data processing RMA normalization was carried out using Partek Genome Suite v6.4.
