Title |
Control 658, fat mRNA |
Sample type |
RNA |
Source name |
fat from female offspring of fathers fed on control chow
Organism |
Rattus norvegicus |
Characteristics |
strain: Sprague Dawley gender: female tissue: fat treatment: father fed standard diet
Treatment protocol |
Fathers fed on HFD vs. control were mated with females on normal chow. The female offspring's fat transcriptomics were analyzed through microarray analysis.
Growth protocol |
At 15 weeks of age, fathers were mated with normal females consuming chow to generate the F1 offspring.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions (Invitrogen).
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45°C on GeneChip Rat Gene ST 1.0 arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Affymetrix Scanner GC3000.
Description |
Gene expression of fat from female offspring of fathers fed standard chow.
Data processing |
RMA normalization was carried out using Partek Genome Suite v6.4.
