Gastrocnemius muscles were harvested from 8-week-old female control (WT) and muscle-specific Sel1L deficient (KO) Mice.

Growth protocol |
Muscle-specific SEL1L (Sel1LMLC) deficient mice were generated by crossing Sel1Lf/f mice with mice expressing Cre driven by the myosin light chain-promoter (MLC), mice that were Cre-negative were used as controls. All mice strains used were on C57BL/6J background and were fed a regular chow diet (13% fat, 57% carbohydrate, and 30% protein; LabDiet 5L0D) with free access to water.

Extraction protocol |
RNA was extracted using RNeasy micro kit (Qiagen) including a column for elimination of genomic DNA as per manufacturer’s instructions. RNA concentration was determined using NanoDrop 2000 UV-Vis Spectrophotometer. The quality and concentration were determined using the RNA 6000 Nano kit on an Agilent 2100 bioanalyser.

Hybridization protocol |
Hybridization and washing of the Affymetrix GeneChip Mouse Gene 2.1 ST peg arrays were performed on a GeneTitan Instrument according to the manufacturer's recommendations (Affymetrix, Santa Clara, CA, USA).

Expression estimates were calculated applying the RMA algorithm in the Bioconductor library 'Oligo' (v1.54.1).
