RNA was isolated from the triceps muscles using peqGOLD TriFast (PeqLab Biotechnologie GmbH) according to the supplied protocol. The cRNA synthesis and microarray hybridizaton was done according to standard protocols applied in the Laboratory of Functional Genome Research Charité - Core Facility. A total of 8 high-density oligonucleotide mouse GENE 1.0 ST arrays (Affymetrix) were used in this study. Samples from each animal (4 Nf1, and 4 wild-type) were applied individually to each array.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on MouseGene1.0ST array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
Array scanning was performed according to the manufacturer's instruction (Affymetrix).
