Treatment protocol |
Diabetes was induced in femal FVB mice by two intraperitoneal injections of 100mg/kg body weight Streptozotocin in 50mM sodium citrate buffer within a one week interval. Mice with a blood glucose level higher than 250mg/dL were considered diabetic and used for mating. The morning with the presence of a vaginal plug was counted as day 0.5.

Extraction protocol |
Placentae were dissected at E10.5, weighted and stored in Trizol. Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotin-labeled samples were generated using the Affymetrix Eukaryotic One-Cycle Target Labeling Assay.

GeneChip arrays were hybridized overnight in the Affymetrix hybridization oven at 42oC. Washing and scanning was performed using the Affymetrix GeneChip Fluidics Station 450.

GeneChips were scanned using the GeneChip Scanner 3000 7G.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0; Affymetrix GCOS software) using Affymetrix default analysis settings and global scaling as normalization method. Statistical evaluation was done with Cyber-T and GeneSpring.
