Mouse blood was collected with Microtainer® (BD) with EDTA and incubated with 3 ml ACK lysis buffer (Gibco) for 5 min at room temperature to lyse red blood cells. PBMC were spun down by centrifugation at 1500 rpm for 10 min at 4℃ and washed twice with phosphate-buffered saline (PBS). RNA of PBMC was isolated by using Direct-Zol RNA Miniprep Kits (ZYMO RESEARCH) according to manufacturer instructions.

Data processing |
We merged these samples in different panels into one dataset for subsequent data normalization. Data normalization similar to that described by the nCounter Advanced Analysis user manual (version 2.0, NanoString Technologies) was applied. Each count was transformed to a log2 scale and then normalized using positive control probes and housekeeping genes identified by geNorm algorithmt to reduce variations. We also calculated means of paired normal mice at weeks 3 and weeks 5 to 7 to scale mice (paired normal, tumor-bearing mice, and recurrence mice) at these two time points.
