Following standard ChIP-seq protocol. Briefly, T cells were treated with MNase to generate approximately 80% mononucleosomes and 20% dinucleosomes. Chromatin from 1 × 107 cells was used for each ChIP experiment, which yielded approximately 100 ng of DNA. Antibodies against histone H3K4me3 (ab8580, Abcam) and H3K27me3 (07-449, Upstate) were used. After reverse cross-linking and purification with Qiagen PCR purification kit, the ChIP DNA fragments were blunt-ended, ligated to the Solexa adaptors, PCR amplified, and sequenced using the Illumina 1G Genome Analyzer.
