Treatment protocol The isolation of satellite cells from the M. rhomboideus of ten female five- and twenty days old piglets and the establishment and validation of two muscle cell pools (pool 5 (P5) or pool 20 (P20))
For proliferation experiments cells from both pools stored in liquid nitrogen were defrosted and permanently cultured at 35°, 37° (control), 39° or 41°C over a period of 72 h in growth medium
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using TRI Reagent (Sigma, Taufkirchen, Germany) according to the manufacturer’s protocol. After DNaseI treatment the RNA was cleaned up using the RNeasy Kit (Qiagen, Hilden, Germany).
Label biotin
Label protocol biotin-labeled cRNAs were prepared according to the standard Affymetrix protocol from 500 ng total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 42 C on Affymetrix Porcine Snowball Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol Scanning using the GeneChip scanner 3000 were performed according to Affymetrix protocols
Data processing Data were analysed with Affymetrix GCOS 1.1.1 software using global scaling to a target signal of 500.
