Treatment protocol |
HMF3A cells were cultured at 34°C ±0.5°C and temperature shift experiments performed at 38°C ±0.5°C. Cells were incubated at 38°C ±0.5°C for 7 days before RNA was extracted. HMF3A cells were serum starved by culturing in DMEM supplemented with 0.25% heat inactivated fetal bovine serum, 2mM glutamine, 100 units per ml penicillin and 100 micrograms per ml streptomycin. Cultures were fed with fresh medium the day prior to RNA extraction and were harvested at no greater than 80% confluence.

Growth protocol |
HMF3A cells and derivatives thereof were maintained in DMEM supplemented with 10% heat inactivated fetal bovine serum, 2mM glutamine, 100 units per ml penicillin and 100 micrograms per ml streptomcyin and cultured in atmosphere of 5% CO2 and 20% O2.
