Title |
nas4x-1 roots + Fe, biological rep2 |
Sample type |
RNA |
|
|
Source name |
nas4x-1, Roots grown under + Fe
|
Organism |
Arabidopsis thaliana |
Characteristics |
tissue: Roots
|
Treatment protocol |
No special treatment has been applied
|
Growth protocol |
Wild-type and nas4x-1 plants were grown in hydroponic solution containing a quarter strength of Hoagland salts supplied with 10µM FeNa-EDTA. The medium was exchanged weekly. Four weeks after germination, plants were divided into two groups, one of them was kept in the same conditions as mentioned above (10µM FeNa-EDTA, + Fe) while the other group was transferred to Hoagland solution without Fe (0 µM FeNa-EDTA, - Fe). Cultivation took place at 21°C/19°C and 16-h-light, 8-h-dark cycles and a light intensity of 150 µmol x m-2 x s-1.

Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted from 100 mg of root and leaf material with the Quiagen RNeasy Plant Mini Prep Kit.
|
Label |
Biotin
|
Label protocol |
5 µg RNA of each sample were processed into biotin-labeled cRNA and hybridized to Affymetrix GeneChip Arabidopsis ATH1 Genome Arrays (Affymetrix, High Wycombe, U.K.), using the Affymetrix One-Cycle Labeling and Control (Target) kit
