Treatment protocol Water stresses on poplars under regime 1, regime 2, regime 3 and regime 4 were imposed by watering 70%, 50%, 20%, and 5% of daily evapotranspiration back into the selected pots, respectively, for up to seven weeks, watering 100%, assigned as regime 0, of daily evapotranspiration were taken as control for the stress treatment. The amount of daily evapotranspiration of each pot was determined by its average daily mass loss in 10 consecutive days before executing the regimes.
Growth protocol The seedlings of Populus euphratica were grown under a natural photoperiod, humidity and temperaturein 5L (30 cm deep) pots with a mixture composed of sand, loamy soil, peat (1:1:1, v/v) in a rain-shelter field in Beijing

Extraction protocol All of the fifth and sixth leaves of a single poplar in each treatment were collected as its RNA sample pool. Total RNA were prepared mainly using CTAB extraction procedure(Chang et al. 1993)

Label protocol Biotinylated cRNA were labeled according to GeneChip IVT Labeling Kit
 
Hybridization protocol Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Poplar Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using GeneChip TM scanner 3000.

Data processing The data were analyzed with Flexarray software package using RMA for normalization and SAM for differently expression genes.
