Embryos were harvested at 8.5 dpc in chilled DEPC-PBS, dissected out of the yolk sac, the allantois was removed, somites counted for the purposes of stage-matching, then flash frozen in liquid nitrogen.

Growth protocol |
Female Cited2+/— mice were fed with Beekay Rat and Mouse Diet (BK002P) between weaning and 6 weeks of age, and then were randomly assigned to specially formulated high-fat or control diets, which were provided ad libitum. At baseline and after 8 weeks on the diet, the mice were weighed and a 6-hour fasting blood sample was collected. Blood glucose and plasma insulin were measured. Following 8 weeks on diet, Cited2+/— female mice were mated with Cited2+/— males, and the diet continued through pregnancy.

Data processing |
Data generated from scanning was imported into Illumina BeadStudio version 3.2.6 to generate signal values for each bead type. These data were exported to the R statistical computing language where it was background corrected, and transformed and normalised using the BioConductor package, vsn2 (Huber et al. 2002).
