Growth protocol MN cells tumor cells were cultured in Anne-Kelso DMEM supplemented with 20% foetal calf serum, 0.1mM L-asparagine, 100 U/mL penicillin, 100 mg/mL streptomycin, at 37°C and 10% CO2.

Extraction protocol Genomic DNA was extracted from cells using DNeasy Blood & Tissue Kit (Qiagen).
Sequencing libraries were prepared by nested PCR method to amplify integrated CRISPR guides.

Data processing The RNA sequencing reads were trimmed using cutadapt (v1.16.5) to remove poor quality ends.
The resulting reads were aligned and statistical significance was calculated using MAGeCK algorithm (v0.5.8.1).
