MN cells tumor cells were cultured in Anne-Kelso DMEM supplemented with 20% foetal calf serum, 0.1mM L-asparagine, 100 U/mL penicillin, 100 mg/mL streptomycin, at 37°C and 10% CO2.

Genomic DNA was extracted from cells using DNeasy Blood & Tissue Kit (Qiagen). Sequencing libraries were prepared by nested PCR method to amplify integrated CRISPR guides.

Data processing |
The RNA sequencing reads were trimmed using cutadapt (v1.16.5) to remove poor quality ends. The resulting reads were aligned and statistical significance was calculated using MAGeCK algorithm (v0.5.8.1). Genome_build: mm10 (GRCm38) Supplementary_files_format_and_content: AG636_CRISPRscreenCounts_normalised.txt: Normalized count file representing number of reads per guide scored in T0, T10 and T24 samples.
