Treatment protocol skeletal muscles (pectoralis) were collected from broilers and layers at E10, E12, E14 and E18 stages, immediately frozen in liquid nitrogen, and stored at -80°C until RNA isolation.
Growth protocol Meat-type broiler eggs (commercial Arbor Acres) and egg-type layer eggs (White Leghorn) were incubated at 37.5°C.
Extracted molecule total RNA
Extraction protocol total RNA were isolated from skeletal muscles (pectoralis) collected from broilers and layers at the embryonic stages E10, E12, E14 and E18 with TRiZol reagent.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2 ug total RNA.
 
Hybridization protocol Affymetrix Gene Chip array hybridization was carried out according to Affymetrix Expression Analysis Technical Manual by GeneTech Biotechnology Limited Company (Shanghai, China). Following labeled cRNA fragmentation, 15 μg of cRNA were hybridized for 16 hr at 45℃ on Affymetrix Chicken Genome Array. Washing and staining were followed standard Affymetrix protocol.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 7G
