Treatment protocol none
Growth protocol Plants were grown at 18hr day cycles at 60% humidity. Male sterile lines were pollinated by the appropriate pollen depending on the cross, and siliques harvested in liquid nitrogen at 5 days after pollination (DAP). For the msi1 sample, the msi1 mutation was introgressed in a A9 barnase male sterile line, siliques were left to elongate in absence of fertilization and were harvested 7 days after flower opening.

Extraction protocol Total RNA from siliques was extracted using the RNeasy Plant Mini Kit (Qiagen) and concentrated using an RNeasy MinElute Cleanup Kit (Qiagen).
