Extraction protocol |
Collections of 19-20h old Drosophila embryos were collected and dissociated. Cells were stained with Dapi and DRAQ5 to exclude debris. Single cells were sorted into 96-well-plates in Smart-seq2 lysis buffer. Full-length cDNA from individual cells were produced following the SmartSeq2 protocol. Smart-Seq2 (modified from Picelli et al., Nature Protocols 2014) Tagmentation using Homemade Tn5 (Hennig et al., G3 2018)
