Sample type |
RNA |
Source name |
2mm skin biopsy
Organism |
Homo sapiens |
Characteristics |
tissue: Human skin grafted in diabetic mice disease state: diabetic time: 0 hours
Treatment protocol |
Experimental diabetes was induced by intraperitoneal injections of STZ, administered on five consecutive days. Wound healing experiments were performed using a biopsy punch (Pfm Medical Ag.,Cologne, Germany) in the stable human skin engrafted on immunodeficient mice exposed to sustained hyperglycaemia for 6 weeks. Specifically, 2mm-circular excisional wounds were created and the excised tissue was harvested and used as a reference for gene expression analysis
Growth protocol |
Human skin biopsies of healthy donors, isolated by biomechanical and enzimatic digestion and expanded in vitro
Extracted molecule |
total RNA |
Extraction protocol |
RNA isolation was performed using Trizol (Invitrogen) extraction based methods, followed by purification in columns (Qiagen).
Label |
Biotin
Label protocol |
generate double-stranded cDNA using the protocol for one-cycle cDNA synthesis. Biotin-labeled cRNA was fragmented and hybridized overnight to Affymetrix HU133 2.0 GeneChip
Hybridization protocol |
16h of hybridization at 45ÂșC,
Scan protocol |
arrays were washed, stained with streptavidin phycoerythrin, and were then scanned with the GeneArray Scanner (Affymetrix) to generate an image file.
Data processing |
PCA on samples detected two outliers (C0_04 and C0_06). Raw data from microarrays excluding outliers was normalized using the rma algorithm from the affy package (Bioconductor)
