Treatment protocol |
Neurons were dissociated and microsphere-labeled CSMN, CPN, or CTPN were purified by fluorescence-activated cell sorting (FACS) directly into RNAlater.

Growth protocol |
All neuronal subtypes were purified from C57BL/6 mice. Neuronal subtypes were retrogradely labeled with green fluorescent microspheres injected into appropriate axonal fields: sensorimotor cortex (CSMN and CPN) or visual cortex (CTPN).

RNA was amplified according to the Affymetrix small sample protocol, using two consecutive rounds of linear in vitro transcription to obtain 15-20 ug of amplified and labeled cRNA for each hybridization.
