Treatment protocol |
Bacterial cultures were treated with 0, 1 and 4 µg/ml, respectively, of fosfomycin. Bacterial cells were harvested at the start of the experiment (t0) and at 3 time points (10, 20 and 40 minutes) after treatment. The experiment was repeated at least three times.

Growth protocol |
Staphylococcus aureus, strain ATTC 29213 was used in the experiments. Overnight culture was set in a small volume of cation-adjusted Mueller–Hinton broth medium (Sigma-Aldrich) and grown in Erlenmeyer flask on gyratory shaker (200 rpm) at 37°C. The overnight culture was diluted 100-fold in 300 ml of medium and grown under same conditions in 1-L Erlenmeyer flasks until OD600 (measured by Beckman DU640 spectrophotometer) reached 0.3, which corresponded to the early exponential stage of the growth curve.

RNA was isolated from an aliquot of 7ml of the culture using enzyme cell wall lysis (30) followed by RNeasy Mini Kit (Qiagen) purification.
