Aerial parts of 4 weeks plants were collected and immediately placed on liquid N2 and ground using a mortar and pestle. RNA was extracted using Direct-zol kit (Zymo Research) according to manufacturers instructions. Total RNA integrity was analyzed on Agilent 2100 Bioanalyzer. 10ug of RNA was resolved on denaturing 15%PAA gel and small RNA fraction was cout out from the gel Libraries were constructed using the TruSeq SmallRNA Library Preparation Kit (Illumina) as described in the manual.
