Twenty WT FVB, WT C57, and B2KO mice were given intraperitoneal injections of avertin (100μl/10g body weight) and sacrificed by cervical dislocation. Half of the mice in each group had undergone fourteen day isoproterenol infusions. RNA was extracted from the mouse hearts using Quiagen RNeasy® Midi kit. Samples were homogenized in buffer containing guanidine isothiocyanate and beta mercaptoethanol. Ethanol was then added to the lysate. The sample was then applied to the RNeasy column. RNA was eluted from the column using 80 ul of RNase-free water.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix)

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip 430-2 Mouse Gene array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Genechip Scanner 3000.
